No, this is in addition to PL.  If a user sends regular PL, then his repeater operats normally.  If he sends APRS data at XX HZ, then this also triggers a UNSQL output to bring up the repeater.  The normal PL and this APRS DPL detector are -ORED- to the repeater transmitter keying...
